Adding spot colour to colour calibration

We are having trouble with a colour not coming out properly on our Epson 10600 Ultra Chrome. It is one pms colour (300c). It comes out way to dark. We are using O.R.I.S Colour Tuner 5.0.6. How do I add a spot colour to the calibration that is already in place? Simplet terms would be greatly appreciated. The guy I normally deal with is away on vacations.
