Go to "Disk Utility" and select the drive you want to fix permissions on. On the bottom left of the dialog box, there should be two buttons, click on the one that says fix (or repair) permissions. I never bother with "check permissions, since I find my permissions are always hosed.

You can also download a really clever freeware app called Onyx that bunches all sorts of essential tasks together under one common interface

I repair permissions almost daily now, since I have lost permissions on fonts and on printers (which intitiated a massive chain reaction of evil - but did uproot the fact that the IT guys used my RIP's IP addy for one of DP's IP printers).
